Karyotype |
This is a hyperdiploid human cell line of male origin with a modal chromosome number of 49 and a 4% polyploidy rate. Most of the cells examined contained the t(11;14)(q13;q32) derivative chromosome common to Mantle Cell Lymphoma. Other derivative chromosomes present in most of the examined cells included: del(5)(p15), der(9)t(9;?), der(14)t(8;14)?, and add(17p). Overall, the karyology was consistent with published data. |

Derivation |
The Z-138 cell line was derived from a patient who initially was diagnosed with chronic lymphocytic leukemia (CLL). About 2 years later, the neoplasm had transformed into an aggressive, mature B-cell acute lymphocytic leukemia (B-cell ALL). According to the criteria of the current World Health Organization lymphoma classification, this neoplasm is best classified as mantle cell lymphoma, with blastoid transformation present in the terminal phase of disease. |

Z-138 cells have a complex karyotype, including the t(11;14)(q13;q32) abnormality, which is common in B-cell neoplasms.

The base medium for this cell line is ATCC-formulated Iscove's Modified Dulbecco's Medium, Catalog No. 30-2005. To make the complete growth medium, add the following components to the base medium:
horse serum to a final concentration of 10%

Subculturing |
Cultures can be maintained by the addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 x 105 to 2 x 105 viable cells/mL. Do not allow the cell concentration to exceed 3 x 106 cells/mL.
Subcultivation ratio: A subcultivation ratio of 1:5 to 1:15 is recommended.
Medium renewal: Add fresh medium every 2 to 3 days (depending on cell density). |
