Restriction digests of the clone give the following sizes (kb): BamHI--9.5; HindIII--9.5; BamHI/HindIII--6.0, 3.3; EcoRI--9.5; PstI--4.2, 1.1, 0.78, 0.66, 0.5. Genomic clone of the entire human involucrin gene, including the 5' untranslated sequence, 2 exons and 1 intron. The coding region consists of 59 repeats of a 30 nucleotide sequence. The 5' flanking regions (to about 730 bp upstream of transcription start site), the 43 bp noncoding exons, and the 1.2 kb single introns are 81% identical in the human and ring-tailed lemur (Lemur catta). The coding region of the common involucrin gene contains 39 repeats of 10 codons. One of these repeats is missing in some individuals, giving rise to the PstI polymorphism. A 6 kb HindIII/BamHI fragment containing the entire gene has been used as a probe to detect informative bands between 0.82 and 0.91 of the PstI polymorphism. |
